Bioactive Compounds Produced by Pseudoalteromonas Affect Marine Biofilm Formation ArchiMer

Bacterial biofilms, frequently in association with algae, protozoa and fungi, are found on all submerged structures in the marine environment. Biofilms are responsible for a range of surface‐associated and diffusible signals, which may moderate the settling behaviour of cells, spores and larvae. Thus, marine microorganisms are a new source of bioactive compounds, which enhance or inhibit the settlement of organisms.

Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Cryopreservation of sperm in marine fish ArchiMer

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Direct and fast detection of Alexandrium minutum algae by using high frequency microbalance ArchiMer

In this paper, a simple detection of a toxic algae, Alexandrium minutum, was developed using highly sensitive quartz crystal microbalance. In terms of performance, compared with other conventional analytical tools, the main interest of our immunosensor is based on a fast and direct detection of these living cells. This system requires the use of one monoclonal antibody directed against the surface antigen of A. minutum. We demonstrate that the whole living and motile algae are caught and detected. The high specificity of the biosensor is also demonstrated by testing several other dinoflagellate species. The frequency shift is correlated to the A. minutum cells concentration. This simple system is potentially promising for environmental monitoring purposes.

The scientific question addressed in this work is: what hides beneath first order kinetic constant k (s(-1)) measured for hybridization of a DNA target on a biosensor surface. Kinetics hybridization curves were established with a 27 MHz quartz microbalance (9 MHz, third harmonic) biosensor, constituted of a 20-base probe monolayer deposited on a gold covered quartz surface. Kinetics analysis, by a known two-step adsorption-hybridization mechanism, is well appropriate to fit properly hybridization kinetics curves, for complementary 20-base to 40-base targets over two concentration decades. Suite à l’événement de grande ampleur d’efflorescence d’Alexandrium minutum et des toxicités associées en Rade de Brest en 2012, un projet a été élaboré avec la Région Bretagne dans le cadre du développement du port de Brest. L’Ifremer a organisé un échantillonnage dans l’estuaire de la rivière de Daoulas, zone la plus touchée en 2012, au cours des printemps-été 2013 et 2014. Un suivi de la présence d’A. minutum dans les sédiments a également été réalisé en hiver 2014. Long-term effects of the cryopreservation of turbot (Psetta maxima) spermatozoa ArchiMer

The survival of turbot eggs and the rearing capacities of larvae stemmed from artificial fertilization practices using frozen-thawed spermatozoa were evaluated. Furthermore, the viability of sperm samples stored during a 9 month period in liquid nitrogen was assessed. No significant difference in the fertilization rate, hatching rate, survival and wet weight of 10-day old larvae were observed using fresh or frozen-thawed spermatozoa. The motility recorded at 10 s and 60 s post-activation and the fertilization capacity of frozen-thawed spermatozoa were not significantly decreased during a 9 month storage period in liquid nitrogen. Marine fish spermatozoa: racing ephemeral swimmers ArchiMer

After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. The cost of such 'hyperactivity' is a very rapid consumption of intracellular ATP that outstrips the supply....

Up to now, toxic species are monitored by expensive and time-consuming laboratory analysis of seawater samples: after a 24h decantation, the samples are observed with an optical microscope and the cell concentration per species is determined. In this context, systems able to detect HAB species directly underwater would be of great help. An assay based on an SPR biosensor was then developed for detecting Alexandrium minutum.

NOUBA Project Novel Natural Molecules Inhibiting the Development of Marine Bacteria in Biofilms ArchiMer

Marine organisms represent a rather unexplored source of new activity and biological functions of molecules for biotechnologies. The research in chemical ecology of the marine environment leads to looking at metabolites of recognition or defence produced by these models. Marine bacteria belonging to the Pseudoalteromonas genus of the Gammaproteobacteria class are often found in association with marine eukaryotes, and their ability to produce a variety of biological activities attracted a particular attention. The dinoflagellates of Alexandrium genus are known to be producers of paralytic shellfish toxins that regularly impact the shellfish aquaculture industry and fisheries. Accurate detection of Alexandrium including A. minutum is crucial for environmental monitoring and sanitary issues. In this study, we firstly developed a quantitative lateral flow immunoassay (LFIA) using super-paramagnetic nanobeads for A. minutum whole cells. This dipstick assay relies on two distinct monoclonal antibodies used in a sandwich format and directed against surface antigens of this organism. No sample preparation is required. Short-term storage and cryopreservation of turbot (Scophthalmus maximus) sperm ArchiMer

Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

Toward in situ detection of algae species ArchiMer

For the last few years there is a more and more pressing need to develop system for detecting HAB at their early stage. Researchers have then been working on new in situ sensor. In this paper a submarine transducer based on surface plasmon resonance is reported. It demonstrated a refractive index resolution of 3.10(-6), which is promising for later phytoplankton detection.
